Polyphenol-Nad+ Nps Inhibit Pdlsc Senescence For Periodontal Regeneration
To develop NAD+-loaded metal-phenolic network self-assembled nanoparticles for enhancing intracellular NAD+ levels and scavenge ROS in periodontal ligament cells, inhibiting cellular senescence, and promoting regeneration of inflammatory periodontal tissue. MPN-NAD NPs were prepared at room temperature via a one-step method. The presence of MPN and NAD+ in NPs was verified by FTIR. NAD+ released from MPN-NAD NPs at pH 5.0 and pH 7.4 were determined by NAD+/NADH detection kits. The surface morphology and element content analysis of the material was detected by SEM and TEM. The radical scavenging ability of the MPN-NAD NPs was performed by using DPPH and H2O2 as model radicals. Human PDLSCs were isolated and extracted, CCK8 assay was used to evaluate the biocompatibility of MPN-NAD NPs. Following H2O2-induced cellular senescence, PDLSCs were treated with MPN-NAD NPs, qRT-PCR and SA-β-gal were performed to detect senescence-associated markers to validate the regulatory effect of MPN-NAD NPs on PDLSCs senescence. MPN-NAD NPs were spherical nanoparticles with controllable particle sizes. They demonstrate pH-responsive sustained release under acidic conditions and exhibit ROS scavenging capacity. The nanoparticles showed biological compatibility and effectively inhibit cellular senescence of PDLSCs. MPN-NAD NPs effectively replenish intracellular NAD+ levels, inhibit cellular senescence in PDLSCs and scavenge ROS, demonstrating potential as a novel therapeutic strategy for periodontitis.
