Houttuynia Cordata Oil And Its Constituents Effects On Pdlscs
The study aimed to explore the effects of Houttuynia Cordata Oil(HCO) and its constituent Methyl Nonyl Ketone(MNK) on the biological characteristics of PDLSCs in inflammatory microenvironment. PDLSCs were isolated using an enzyme digestion method. HCO was extracted by steam distillation. The CCK-8 method was used to determine the safe concentrations of HCO and MNK. An in vitro inflammatory microenvironment was established using 10 ng/mL TNF-α and 5 ng/mL IL-1β. The mRNA expression levels of TNF-α, IL-1β, and IL-6 were detected by RT-PCR. The proliferation ability was assessed by CCK-8 method, cell migration was evaluated by scratch test, and the clonal formation ability was determined by clonal formation test. ALP staining and ARS staining were used to assess osteogenic differentiation. The expression levels of osteogenic-related genes and proteins were detected by RT-PCR and Western Blot. The combination of 5 μg/mL HCO and 50 μg/mL MNK promoted cell proliferation . 10 ng/mL TNF-α and 5 ng/mL IL-1β promoted the expression of inflammatory factors TNF-α, IL-1β, and IL-6 in PDLSCs, inhibited cell proliferation, cell migration and clone formation, down-regulated the expression of ALP, RUNX2, COL I, and OCN, and inhibited the osteogenic differentiation ability of PDLSCs.The intervention of HCO and MNK improved the biological characteristics of PDLSCs with inflammatory microenvironment inhibition. HCO and MNK enhanced the cell activity of PDLSCs, improved their biological characteristics in inflammatory microenvironment, and promoted osteogenic differentiation capacity.
