Anti-Biofilm Effects Of Zoliflodacin On Streptococcus Mutans
To study anti-biofilm effects of Zoliflodacin on conditional cariogenic Streptococcus mutans (S. mutans). Biofilm biomass was quantified by crystal violet assay, colony forming unit (CFU) testing. pH and lactic acid measurement were to monitor acid generation, while anthrone-sulfuric acid method and polysaccharide/ bacterial staining were to detect polysaccharide synthesis. Bacterial activity was revealed by growth curve and live/dead bacterial staining. Quantitative real-time polymerase chain reaction (qRT-PCR) was to detect expression of virulence related genes. Cytotoxicity was evaluated by live/dead cellular staining and CCK-8 assay. Zoliflodacin suppressed S. mutans biofilm formation which was reflected by significantly reduced crystal violet’s absorbance (p<0.05) and CFU (p<0.05). Besides, cariogenic virulence including acid and polysaccharide generation were also decreased observably (p<0.05). Moreover, expression of virulence related genes including gtfB, gtfC and ldh were suppressed significantly (p<0.05). What’s more, Zoliflodacin showed no significant cytotoxicity when compared with its control (p>0.05). Zoliflodacin is potential antibacterial agent in caries control based on its anti-biofilm effects on S. mutans and unconspicuous cytotoxicity.
