The Antibacterial Effect Of Oct-Mpect Hydrogel Against Periodontal Pathogens
To construct a novel drug carrier hydrogel OCT-mPECT and evaluated the the antibacterial effect of it on common periodontal pathogens (F.n, A.a and P.g). OCT-mPECT hydrogel was prepared by nano-coprecipitation technique and evaluated for phase transition behavior, internal structure, rheological behavior, particle size distribution, and drug release curve. Serial plate transfer test (SPTT), confocal laser scanning microscopy (CLSM) and scanning electronic microscope (SEM) were used to evaluate its sustained antibacterial effect against F.n, A.a and P.g. The toxicity of OCT-mPECT to human gingival fibroblasts (hGFs) was determined by CCK-8. OCT-mPECT hydrogel had uniform porous structure, gel-sol phase transition and injectivity. The average nanoparticles size was 115.1nm. OCT was released sustainably and stably for at least 20 days. SPTT showed that the antibacterial effect of Periocline was lower than that of OCT-mPECT at day 3(P<0.05), while OCT-mPECT maintained stable and sustained antibacterial effect at day 14. SEM showed that OCT-mPECT had the least residual bacteria. CLSM showed that the percentage of dead bacteria in OCT-mPECT group was the highest among all experimental groups (P<0.05). OCT-mPECT hydrogel had no effect on activity and proliferation of hGFs (P>0.05). OCT-mPECT is an injectable, sustained-release, and biocompatible drug-loaded supramolecular hydrogel. It can effectively eradicate various periodontal pathogens and disrupted multispecies biofilms through slow drug release, with promising application potential in the local drug therapy of periodontitis.
