Dentin-Cgf Promotes Huvec Proliferation/Migration Via Pi3k/Akt
To assess dentin-concentrated growth factor (dentin-CGF) composites as scaffolds for dental pulp regeneration by examining their effects on HUVEC proliferation/migration and underlying mechanisms. Dentin-CGF (1:1 weight ratio) was immersed in endothelial media to prepare 100/500/1000 mg/mL suspensions. Conditioned media were obtained via filtration. HUVECs were treated with media, and proliferation (CCK-8 assay) and migration (scratch assay) were analyzed. Gene expression (VEGFA, VEGFR2, p110α/β/δ, PIP3, Akt, p27) was quantified by RT-qPCR and WB. Treated cells showed higher OD values (P<0.05) and migration distances (P<0.05) vs. controls. VEGFA, VEGFR2, p110α/β/δ, PIP3, and Akt were upregulated, while p27 was downregulated (P<0.05). Dentin-CGF enhances HUVEC proliferation/migration by activating PI3K/Akt signaling via upregulated PI3K subunits (p110α/β/δ) and downstream effectors (PIP3/Akt). Downregulated p27 indicates cell cycle acceleration, promoting vascular regeneration. This composite is a promising scaffold for pulp regeneration.
