Anti-Inflammatory Nanomaterials Regulating Osteoimmune Microenvironment For Periodontal Bone Regeneration
To develop a bioactive patch regulating the osteoimmune microenvironment for stem cell-based bone regeneration. Biodegradable bioactive nanomicelles (PPT NMs) were synthesized and incorporated into electrospun nanofibers to fabricate a bioactive patch. A dual-membrane bio-patch was engineered by combining the bioactive patch with PDLSC-derived cell sheets. Surface functionalization with tannic acid enhanced its adhesion to bone defects. The nanotherapy significantly improved the osteogenic capacity of periodontal ligament stem cells (PDLSCs) under pathological conditions by dual modulation of the IL-17 signaling pathway and ferroptosis. The dual-membrane patch synergistically enhanced bone regeneration by regulating the osteoimmune microenvironment and promoting stem cell differentiation. Tannic acid functionalization improved adhesion, prolonged local retention, and maintained bioactivity, achieving superior repair in rat mandibular and cranial defect models. This osteoimmune-modulating bio-patch combined with stem cell therapy effectively promotes bone regeneration. The strategy also holds potential for spinal cord injury, wound healing, and burn repair in stem cell-based therapies.
