Ph-Responsive Nanozymes Alleviate Inflammation And Bone Resorption In Periodontitis

This study aims to develop a multifunctional nanozyme for periodontitis therapy by integrating ROS scavenging, macrophage polarization modulation, and antibacterial activity, addressing the limitations of conventional materials in multi-target intervention. Hydrothermally synthesized nanozymes were evaluated for antibacterial activity (using SEM and live/dead staining), ROS scavenging(using DCFH-DA assay staining and qPCR), and osteogenic differentiation(usingALP/ARS staining). Macrophage polarization (M1/M2) was analyzed via immunofluorescence(iNOS/CD206). Periodontitis models in SD rats(4-6w,male) were established to assess tissue regeneration and inflammatory response. The nanozymes effectively inhibited bacterial growth, scavenged ROS, reduced pro-inflammatory macrophages, and enhanced anti-inflammatory macrophages. In vivo results demonstrated alleviation of inflammation and robust bone regeneration. The nanozymes synergistically remodel the periodontal microenvironment through antimicrobial, immunomodulatory, and osteogenic mechanisms, offering a novel multi-functional strategy for periodontitis therapy.
