Mechanism Of Parallel Topological Morphology Promoting Neurogenic Differentiation Of Hbmscs
In this study, parallel topography electrospinning nanofiber scaffolds were used to simulate the neural regeneration induction microenvironment, and parallel topological topography was used to study the induction of human bone marrow-derived mesenchymal stem cells. The potential of hBMSCs to differentiate into neural-like cells and the specific mechanism were identified. Parallel topography polylactic acid electrospun scaffolds were fabricated using electrospinning technology. The neurogenic differentiation potential of hBMSCs induced by parallel topography electrospinning was evaluated through quantitative real-time PCR and immunofluorescence techniques. hBMSCs were seeded onto parallel topography electrospun scaffolds, smooth membrane groups, and neurogenic induction medium groups, respectively. The expression levels of key neurogenic differentiation marker genes were detected by quantitative real-time PCR. miRNA microarray sequencing was performed to enrich significantly differential miRNAs and analyze related pathway changes. The results demonstrated that parallel topological topography effectively promoted neurogenic differentiation of hBMSCs. Concurrently, miRNA microarray sequencing revealed a significant increase in miR-451a expression and activation of neurogenic-related pathways, indicating that parallel topological topography promotes hBMSC neurogenic differentiation by enhancing miR-451a expression. Parallel topological morphology can promote neurogenic differentiation of hBMSCs by increasing the expression of miR-451a. This discovery provides a new idea and method for the application of neural tissue engineering seed cells, and provides a new approach and theoretical foundation for the development of long-term stable and functional tissue engineering nerves.
