Cibinetide Alleviate The Progression Of Apical Periodontitis Via Sirt1 Axis
This study aims to examine the protective benefits and underlying mechanism of Cibinetide in alleviating pro-inflammatory responses of macrophages and osteoclasts differention in AP. Firstly, to evaluate the aberrant activation of SIRT1/NF-κB/IL-1β signaling pathway in periapical lesions and the role of Cibinetide, forty male C57BL/6 mice, aged 6-8w, were randomly divided into three groups: Control (Cont) group(n=20), apical periodontitis (AP) group(n=10)and AP+Cibinetide group(n=10). This study received approved from the Laboratory Animal Ethics Committee of Xi’an Jiaotong University (XJTUAE2024-2603). Thereafter, SIRT1/NF-κB/IL-1β was blocked by selisistat, and the changes in inflammation levels and bone resorption volume in periapical lesions were assessed. The student’s t-test and analysis of variance (ANOVA) was utilized for multiple groups to ascertain statistical significance. A p-value below 0.5 was deemed indicative of statistical significance. In vivo AP model revealed a considerable reduction in SIRT1 expression in periapical lesions, while the expression of acetylated NF-κB (p65) and IL-1β was notably down-regulated, which can be reversed by treatment of Cibinetide. Furthermore, the administration of Cibinetide in AP mouse may effectively diminish inflammatory infiltration, the quantity of osteoclasts, and the volume of bone loss in the periapical region. The treatment of Cibinetide in vitro has the same results. Cibinetide exerts a protective role via regulating the SIRT1/NF-κB/IL-1β signaling pathway in apical periodontitis
