The Effects Of Bmsc-Derived Exosomes On Oxidative Stress
To explore the protective effect of exosomes from different sources of mesenchymal stem cell on BMSCs under oxidative stress. BMSCs-EXO and BMSCs-EXO were extracted from BMSCs of P3-P5 generation, respectively. BMSCs were pretreated with 300umol/L hydrogen peroxide to construct a model of oxidative stress. After the successful construction of the model, they were divided into groups. Control group: BMSCs pretreated with hydrogen peroxide; Experimental group A: BMSC pretreated with hydrogen peroxide plus BMSCs-EXO; Experimental group B: BMSCs-EXO after BMSCs pretreated with hydrogen peroxide and osteogenic induction. By cck-8, ROS, fluorescent probe, SOD, mitochondrial membrane potential, qRT-PCR, Western blot and ALP staining, alizarin red staining, the effects of different sources of MSCS exosomal BMSCs on cell viability, intracellular ROS level, osteogenic differentiation and iron death were investigated. After the intervention of the two exosomes, ROS levels were significantly decreased, and the expressions of NRF2, CAT, SOD and other antioxidant markers were promoted (P<0.05) and the expressions of ALP, alizarin red and osteogenic markers were increased (P<0.05).Promote the expression of negative iron death related factors such as GPX4 and XCT and inhibit NCOA4 expression (P<0.05). BMSCs-EXO and osteogenic induction of BMSCs-EXO can reduce the death levels of reactive oxygen species and iron in BMSCs under oxidative stress, and promote the osteogenic differentiation of BMSCs under oxidative stress.
