Ldp Immobilization On Psbma-Peg Brush: Mechanisms And Implant Integration
Although titanium implants have been applied in dental clinics to replace lost teeth and to restore masticatory function for decades, strategies to design the surface of the transmucosal sites of implants to achieve ideal and predictable biological sealing following implantation remain to be optimized. In the present study, we introduced a hydrophilic polysulfobetaine-polyethylene glycol (PSBMA-PEG) polymer brush covalently linked with LDP on the permucosal implant surface to promote soft tissue integration surrounding implants. Firstly, we grafted PSBMA-PEG polymer brushes onto the transmucosal segment of implants and covalently linked LDP, creating a specific PSBMA-PEG-LDP coating. Then we assessed its biological impact on human immortal keratinocytes (HaCaTs) and Mϕs. Using a rat maxillary immediate implant model, we further explored the influence of the coating on soft tissue integration and Mϕ polarization in vivo. Finally, we delved into the mechanisms underlying the coating's ability to bolster cellular attachment and gingival epithelial closure. LDP could be grafted to the PSBMA-PEG polymer brush in a stable manner. PSBMA-PEG-LDP coating improved HaCaTs attachment and promoting the expression of M2 phenotype, as evidenced by morphological and immunofluorescence analyses. PSBMA-PEG-LDP-Ti induced better biological sealing at transmucosal sites following immediate implantation into Sprague-Dawley rats through activating Integrin/FAK/PI3K/AKT signaling pathway. The present study demonstrated that the polymer-tethered peptide coating could be used to enhance peri-implant tissue attachment to titanium implants by functioning as a transmucosal barrier.
