Salvia Miltiorrhiza Impacts Wnt/B-Catenin In Fluoride-Affected Orthodontic Rats?

In this study, we investigated the effect of salvia miltiorrhiza on the Wnt/β-catenin pathway in the orthodontic rats with fluorosis. 100 3-week-old male SD rats were divided into five groups: orthodontic (O), fluoride-orthodontic (FO), and high, medium, low salvia intervention fluoride-orthodontic (SH,SM,SL) groups. Each group had four time points (3, 7, 14, 21 days) based on tooth movement cycles. The group O drank tap water (fluoride content<0.08 mg/kg), while others received 150 mg/L NaF solution. After 3 months, orthodontic devices were installed . Salvia groups gavaged salvia suspension: SH (2.16 mg/g), SM (1.08 mg/g),SL (0.54 mg/g). Samples were taken for RT-qPCR and Western-Blot to detect Wnt3a and β-catenin expression. Compared with group O, the expression of Wnt3a and β-catenin decreased in group FO . The expression of them increased to different degrees in each group after salvia miltiorrhiza intervention. Both group SM and SH had a restorative effect in the early stage and also promoted overexpression exceeding those of the orthodontic group, whereas the group SL promoted overexpression in the late stage of tooth movement. (P < 0.05). Salvia miltiorrhiza can affect orthodontic tooth movement by promoting the expression of Wnt/β-catenin pathway, and there is a certain time and dose dependence of the intervention effect, and salvia miltiorrhiza middle and high concentrations have the most significant restoring effect in the pre-movement period of teeth.
