Resveratrol Photodynamic Therapy System For The Treatment Of Periodontal Disease
In this study, we proposed to self-assemble resveratrol (Res) photodynamic nanoparticles with PEG hydrogel to prepare an injectable photodynamic drug delivery system for convenient clinical application and firm adhesion and sustained release in periodontal pockets. The present study aims to investigate the dynamic process of the drug delivery system and the mechanism of action of Res, and to provide a new basis for the pharmacological treatment of patients with diabetic periodontitis from the perspective of regulating mitochondrial autophagy. We combined carboxymethylchitosan (CMCS) with indocyanine green (ICG) to form photoactive nanoparticles and encapsulated Res with poly(ethylene glycol)-based (PEG) hydrogel to form a novel injectable photodynamic self-assembled hydrogel system (CMCS@Res@ICG-PEG). Through a series of material characterisation and in vivo/in vitro experiments, it was confirmed that CMCS@Res@ICG-PEG could achieve efficient bactericidal effect at the initial stage after drug administration by photodynamic force; and regulation of mitochondrial autophagy by sustained release of Res. The hydrogel system modulates the immune-inflammatory response of the local periodontal microenvironment through photodynamic synergism to improve inflammation in patients with diabetic periodontitis.
