Bactericidal Effect Of Paw Generated By A Novel Potable Device

To address the limitations of traditonal plasma devices (bulky size, professional operation requirements) and the reduced antibacterial effect of plasma activated water (PAW) in the presence of organic matter, a novel super-portable plasma device was applied and the low concentration of H2O2 was added to enhance the bactericidal effect of PAW in the organ-rich oral environments. A super-portable plasma device weighted only 1 kg was applied to generate PAW. Staphylococcus aureus, Escherichia coli and Porphyromonas gingivalis were selected as the test bacteria. CFU counting and bactericidal rate were employed to evaluate the bactericidal effect of PAW. OES was used to identify the major reactive species in plasma jet. Different concentration of H2O2 was added to PAW (H2O2/PAW), the pH, ORP, conductivity, H2O2 and NO content of H2O2/PAW were measured to explore the bactericidal mechanisms. CCK-8 assay was used to assess the cytotoxicity of 0.1% H2O2/PAW on human periodontal ligament fibroblasts. Thenovel plasma device generated PAW that achieved 98.94% (10 min) and 99.66% (20 min) bactericidal rates against S. aureus. PAW was effective against S. aureus, E. coli, and P. gingivalis. Adding 0.1% H2O2 significantly improved PAW's antibacterial effect in BSA presence (p<0.05), by decreasing pH, increasing ORP, conductivity, H2O2, and NO levels. Cytotoxicity assessment showed 0.1% H2O2/PAW had acceptable biosafety. PAW generated by the portable device exhibited excellent bactericidal effects. Adding 0.1% H2O2 maintained its efficacy in organic-rich environments with acceptable biosafety.
