1,25(Oh)2D3 Reverse Lps-Induced Osteogenesis In Hpdlscs
The present study aims to evaluate effects of 1α, 25-dihydroxyvitamin D3 (1,25(OH)₂D₃) on human periodontal ligament stem cells (hPDLSCs) osteogenesis in an inflammatory setting and to investigate the underlying mechanisms. The research was approved by the Ethics Committee. Using the tissue block method to obtain hPDLSCs from extracted premolars or third molars which were collected from individuals aged 14 to 23 years. hPDLSCs were cultured in osteo-inductive medium with 1,25(OH)2D3 and lipopolysaccharide (LPS) in combination or alone. The impact of 1,25(OH)2D3 on the osteogenic differentiation of hPDLSCs was investigated by alizarin red and ALP staining. The expression levels of osteogenesis-related genes, TAZ as well as the activation of NF-κB signaling pathway in hPDLCs during osteo-inductive culture were assessed by western blot and real-time quantitative PCR(qRT-PCR). LPS inhibited osteoblastic differentiation in hPDLSCs. And 1,25(OH)2D3 reversed the negative effects of LPS on osteogenic differentiation and increased the expression of osteogenesis-associated genes and TAZ. For mechanisms, 1,25(OH)2D3 could block the NF-κB signaling pathway by inhibiting p65 phosphorylation to upregulate TAZ and reverse the LPS-induced inhibition of osteogenic differentiation. 1,25(OH)2D3 reverses LPS-induced inhibition of osteogenic differentiation by promoting TAZ expression through inhibiting the NF-κB signaling pathway.
