Role Of SRSF3 In Aging Periodontal Ligament Stem Cells
Alternative RNA splicing generates multiple mRNA isoforms by combining different splice sites, encoding proteins with diverse functions and expanding protein expression diversity. Aging in periodontal tissues impairs their basic functions, leading to increased tissue destruction and impaired regeneration. Changes in alternative splicing are closely linked to aging. Currently, the alterations in mRNA splicing patterns and the expression levels and functions of splicing factors during periodontal tissue aging remain poorly understood. In this study, we established an inflammatory senescence cell model and used techniques such as RNA interference (RNAi), RNA sequencing (RNAseq), chromatin immunoprecipitation (ChIP), and lentivirus transduction to investigate the biological function, signaling pathways, and potential molecular mechanisms of the splicing factor SRSF3 in the inflammatory senescence process of periodontal cells. We found that the splicing factor SRSF3 acts as a regulator of inflammatory senescent periodontal ligament stem cells by influencing various alternative splicing events, promoting the progression of senescence. Specifically, we discovered that SRSF3 affects the alternative splicing of important regulatory molecules, such as exon 8 of SIRT1, promoting the generation of SIRT1 isoforms that lack exon 8 and accelerating senescence. This project reveals the significant role of SRSF3 in the occurrence and development of age-related periodontitis and suggests it as a potential therapeutic target for age-related periodontitis.
