In Vitro Therapeutic Effect Of Win-55 On TMJOA

This study evaluates the anti-inflammatory effects of WIN-55,212-2 mesylate in an IL-1β-induced inflammatory model of rat temporomandibular joint (TMJ) condylar chondrocytes, aiming to provide new insights for treating temporomandibular joint osteoarthritis (TMJOA). Condyle chondrocytes were extracted from SD rats, and an in vitro inflammatory model was established using IL-1β. Cells were treated with various concentrations of WIN-55. Inflammatory-related gene expression (Il1β,Il6,Tnfα, etc.) was detected using quantitative real-time PCR, while ELISA was used to assess inflammatory protein levels. CCK-8 assay and fluorescence staining were applied to evaluate cell proliferation and toxicity. WIN-55 significantly inhibited the expression of inflammatory factors and reduced the secretion of inflammatory proteins at concentrations of 5μM-10μM, without noticeable cytotoxicity. WIN-55 demonstrated a significant anti-inflammatory effect in the in vitro TMJOA model, offering a novel therapeutic approach for TMJOA. Further studies are needed to elucidate its underlying mechanism of action.
