Effect Of Tweak On Orthodontic Tooth Movement And Potential Mechanisms
To establish an orthodontic tooth movement (OTM) model and observe the effects of different concentrations of TWEAK on the rate of tooth movement, thereby identifying the optimal concentration of TWEAK for influencing OTM. Subsequently, the effects of TWEAK and TWEAK combined with low-level laser (LLL) on periodontal tissue remodeling and macrophage polarization will be investigated to preliminarily explore the underlying mechanisms. Furthermore, transcriptomic data analysis will be performed to identify differentially expressed genes, aiming to elucidate the biological functions and regulatory mechanisms of TWEAK during orthodontic tooth movement. This study aims to provide a new theoretical basis for accelerating orthodontic tooth movement and shortening treatment duration. Thirty 8-week-old male Sprague-Dawley (SD) rats were selected, and an orthodontic tooth movement model was established. A 60 μL solution of TWEAK at different concentrations was microinjected into the periodontal ligament of the bilateral maxillary first molars. H&E staining, MASSON staining, TRAP staining, Immunofluorescence staining, transcriptomic sequencing analysis was used to observe the expression of periodontal remodeling-related factors, macrophage polarization subtypes and the mechanism by which TWEAK influences OTM. 1.The combined application of TWEAK and LLL promotes periodontal tissue remodeling and macrophage polarization during orthodontic tooth movement; 2.TWEAK may enhance the speed of orthodontic tooth movement (OTM) and periodontal remodeling by influencing motor proteins. Within a certain dose range, TWEAK promotes the speed of orthodontic tooth movement.
