Multi-Omics Analysis Of Effects Of Simulated Microgravity In Hdpscs

To investigate the effects of simulated microgravity in human dental pulp stem cells (hDPSCs) . hDPSCs were isolated from third molars of healthy volunteers aged 19-29 years. Phenotypic analysis was carried out by flow cytometry, and proliferation was assessed with the CCK-8 assay. Transcriptomic and proteomic analyses were employed at 3 day and 5 day respectively to investigate the biological effects and molecular mechanisms of hDPSCs under simulated microgravity environment. Simulated microgravity suppresses the proliferative capacity of hDPSCs, associating with alterations in biological processes including DNA transcription and cell cycle regulation. Osteogenic differentially expressed proteins were found to participate in key biological processes, including extracellular matrix formation, regulation of cell protrusion extension, and inhibition of cell growth, activating cytoskeletal metabolism, extracellular matrix receptor interactions, and critical signaling pathways such as HIF-1 (hypoxia-inducible factor-1) and oxygen-sensing mechanisms. The simulated microgravity environment induces premature cellular senescence in hDPSCs. Integrated multi-omics analysis revealed that the HIF-1 signaling pathway may serve as a key regulatory mechanism in response to microgravity.
