Dual-Layer Microneedling Promotes Regeneration Of Diabetic Periodontal Tissues

Design of a multifunctional dual-layer microneedle for regeneration of periodontal soft and hard tissues in diabetic patients[1]. [1] Qu Y, Zeng H, Wang L, et al. Microenvironment‐Regulated Dual‐Layer Microneedle Patch for Promoting Periodontal Soft and Hard Tissue Regeneration in Diabetic Periodontitis. Advanced Functional Materials, 2024, 35(13): 2418076-2418094. The morphology, mechanical properties, degradation behavior, and reactive oxygen species (ROS)-scavenging capacity of double-layered microneedles (d-MNs) were characterized. The cytotoxicity of d-MNs was assessed, along with their osteogenic, angiogenic, ROS-eliminating, and M2 macrophage-polarizing capabilities. A diabetic periodontitis model was established using male Wistar rats, which were then treated with d-MNs for 14 and 28 days. Histological analyses including H&E staining, PAS staining, Masson staining, immunohistochemical staining, and micro-CT scanning were performed on tissue samples, followed by comprehensive data analysis. The animal experimental procedures were approved by the Animal Ethics Committee of Lanzhou University (LZUKQ-2023-050). The d-MNs demonstrated excellent structural and mechanical properties, showed efficient gum penetration and biodegradability (>90% in 28 days), maintained high cell viability (>85%), and exhibited multi-functional therapeutic effects including ROS-scavenging, promoted osteogenesis and angiogenesis, induced macrophage polarization to M2 phenotype, reduced inflammation (decreased IL-6/TNF-α), enhanced tissue regeneration (increased VEGF/RUNX2/COL-Ⅰ), and significantly improved alveolar bone repair. The novel d-MNs offer a transformative therapeutic strategy for diabetic periodontitis, with excellent prospects for clinical implementation in modern dentistry.
