Self-Feedback Hydrogels For The Treatment Of Diabetic Peri-Implantitis

To improve the therapeutic effect of diabetic peri-implantitis via a magnesium releasing self-feedback hydrogel, exploring its inhibition on the oxidative stress of macrophages and clarifying new mechanism of Mg2+ in this process. Self-feedback magnesium-releasing hydrogels will be synthesized and characterized by 1H-NMR, FT-IR and UV-vis and further evaluated on their ROS responsiveness and the controlled releasing curves of magnesium ions by ICP-AES. The oxidative stress regulation effect will be assessed in vitro by using BMDM cells via RT-qPCR, WB flow cytometry and fluorescence imaging, and the specific targets of magnesium ions on mitochondria will be explored by TEM, fluorescence imaging and RT-qPCR to reveal the mechanism by which they inhibit NLRP3 inflammasomes. Finally, the anti-inflammatory and bone restoration effect of the hydrogel will be evaluated in vivo, using diabetic peri-implantitis model in SD rats via biochemical analysis, histological sections, immunohistochemistry and micro-CT, which has been approved by the ethics committee. 1. The hydrogel can absorb excessive ROS, limiting the oxidative stress of macrophages in a certain range via the controlled release of Mg2+. 2. Mg2+ can block mitochondrial Ca2+ overload via competitive inhibition on the mitochondrial calcium uniporter, so as to compromise NLRP3 inflammasome activation, reduce oxidative stress and antagonize M1 polarization. 3. The hydrogel can alleviate inflammation and promote bone restoration in vivo. The self-feedback hydrogel can regulate the oxidative stress of macrophages to promote bone restoration, effectively improving the prognosis of diabetic peri-implantitis.
