Pdcd4 Played A Vital Role In Odontogenic Differentiation Of Hdpscs
To clarify the role of PDCD4 in development of dental embryo and the odontogenic differentiation of hDPSCs, further providing a feasible target for repair of dental pulp injury. HE and immunohistochemical staining methods were used to verify the expression of PDCD4 in dental embryonic tissues during different periods (E14.5, E16.5, E18.5, P1 and P7). Also, stable cell lines shPDCD4-hDPSCs were constructed successfully. And RT-PCR, alizarin red staining were used to verify the effect of PDCD4 in the odontogenic differentiation of dental pulp stem cells. The statistical method used was t test, and p<0.05 represented statistical significance. The results showed that PDCD4 was expressed before mineralization stage of hard tissue. It was mainly expressed in the inner and outer enamel epithelium and dental papilla at the stage of early bell, while expressed in odontoblast cell layer and surrounding dental papilla cells at the late bell period, suggesting that PDCD4 may play a certain role in early stage of tooth embryo mineralization. Meanwhile the expressions of DSPP, DMP-1 and Runx2 genes related to odontogenic differentiation were both significantly increased after odontogenic induction of ShPDCD4-hDPSCs. Both qualitative and quantitative analysis of alizarin red staining showed a significant increase in mineralized nodules. PDCD4 may play a role in the odontogenic differentiation of dental pulp stem cells.
