Rapid Construction Of Dualfunctional Coating For Denture Liners Against Stomatitis

This study aims to develop a bench-to-chairside approach for preventing denture stomatitis via rapid construction of anti-adhesion and antimicrobial coating on denture liners, which achieved by grafting the zwitterionic polymer 2-Methacryloyloxyethyl phosphorylcholine (MPC) incorporated N-halamine antibacterial functional groups. Uniformed silicone soft liner materials (SiR) were immersed into the prepolymer solution and initiated by UV light for grafting polymer coating onto the SiR surface, SiR-P(MPC-AMH). Sodium hypochlorite solution was used to convert the amine groups in the 2-aminoethyl methacrylate polymer into N-halamine functional groups, SiR-P(MPC-AMH)/NCl. The morphological features, functional groups, hydrophilicity, roughness, and changes in mechanical properties of the coatings were characterized. The biocompatibility, anti-adhesion and antimicrobial properties of coatings were evaluated. (1) SiR-P(MPC-AMH)/NCl exhibited excellent superhydrophilic properties, lower surface roughness and stable mechanical properties when coating on the denture liners. (2) SiR-P(MPC-AMH) and SiR-P(MPC-AMH)/NCl exhibited minimal impact on the proliferation and cytoskeletal integrity of L929 cells and inflammatory response in the in vivo models, confirming their biocompatibility. (3) SiR-P(MPC-AMH)/NCl inhibited the biofilm formation, and showed excellent anti-microbial efficacy against Staphylococcus aureus and Candida albicans on organotypic and orthotopic infection model. Constructing dual-functional coating materials via grafting MPC incorporated N-halamine antibacterial functional groups on denture liners, augments the anti-adhesion and antimicrobial properties, providing a bench-to-chairside approach for preventing denture stomatitis.
