Mgf Loaded Pcl/Plga Electrospun Nanofibrous Membrane Promoted Periodontal Regeneration

To evaluate the efficacy of a polydopamine(pDA)-coated mechano growth factor(MGF) loaded polycaprolactone/poly (lactic-co-glycolic acid)(PCL/PLGA) electrospun nanofibrous membrane in promoting periodontal regeneration. A PCL/PLGA electrospun nanofibrous membrane was fabricated, followed by pDA coating and MGF loading. Physicochemical characterization included SEM, XPS, AFM and contact angle measurements. Biocompatibility was evaluated using RAW264.7 macrophages and human periodontal ligament stem cells(hPDLSCs) through live/dead staining. Anti-inflammatory and antioxidant effects of MGF were assessed in LPS-induced models via immunofluorescence staining, flow cytometry, and malondialdehyde quantification. The osteoinductive capacity of MGF on hPDLSCs under inflammation were evaluated through Alizarin Red staining and qRT-PCR. In vivo, 52 male Sprague-Dawley rats (6-8 weeks old) were randomly divided into four groups (control, PCL/PLGA, PCL/PLGA@pDA, and PCL/PLGA@pDA-MGF) with surgically created alveolar bone defects with approval from the Animal Ethics Committee. Outcomes were evaluated at 4 weeks post-surgery via Micro-CT scanning and histological staining. Data were analyzed using Image J and GraphPad Prism. P< 0.05 was considered statistically significant. The PCL/PLGA@pDA-MGF membrane revealed favorable hydrophilicity, biocompatibility, and excellent anti-inflammatory and antioxidant properties. MGF promoted hPDLSCs osteogenic differentiation in vitro. In vivo, accelerated bone formation and reduced inflammation were observed 28 days post-surgery. The PCL/PLGA@pDA-MGF membrane demonstrates great potential as a bioactive scaffold for periodontal regeneration by modulating inflammation and promoting osteogenesis, offering a novel strategy for defect repair.
