Construction And Molecular Characterization Of Rare Salivary Gland Tumor Organoids

To establish a personalized research model for rare subtypes of salivary gland tumor, bridging foundational investigations and clinical applications to advance precision oncology in this understudied field. Sample Collection Rare salivary gland tumor tissuescollected under sterile protocols. Ethics: Approved by IRB of China Medical University Hospital of Stomatology (#2025010). Clinical Data Integration Statistical analysis of clinical metadata and organoid parameters. Organoid Histological Evaluation Assessment: Inverted microscopy, H&E/immunofluorescence staining (lineage-specific markers). Morphometric/molecular comparisons to parental tumors using non-parametric tests. Transcriptomic Profiling RNA-seq: Tumor tissues, normal glands, and organoids. Bioinformatics: Dimensionality reduction, consensus clustering (Consensus Cluster Plus), differential expression (DESeq2). 1.Organoid models for six rare salivary gland tumor subtypes—including two benign (myoepithelioma [ME] and canalicular adenoma [CA]) and four malignant (epithelial-myoepithelial carcinoma [EMC], myoepithelial carcinoma [MC], secretory carcinoma [SC], and nonspecific adenocarcinoma [NOS])—were successfully established alongside normal gland organoid controls, demonstrating rapid maturation by day 14 and growth plateauing at day 21. 2.Organoids derived from myoepithelial-dominant tumors (ME, MC) showed reduced proliferation efficiency, while others exhibited robust expansion. 3.These organoids faithfully retained parental tumor morphology, transcriptional profiles and cellular heterogeneity mirroring diagnostic and structural features of original tumors. Salivary gland tumor organoids serve as high-fidelity in vitro models for studying rare salivary gland tumors, effectively recapitulating the morphological and molecular characteristics of their parental tumors offering a powerful platform to advance personalized therapeutic strategies and mechanistic studies for rare salivary gland carcinomas.
