Application Of Plga/Ksl-W Antibacterial Nano-Fiber Membrane
With the continuous development of endodontic theory and treatment technology, microsurgery, as the ultimate defense in preserving natural teeth, emerges as the times require. And regenerative techniques (such as guided bone/tissue regeneration) are increasingly applied to the treatment of large periapical bone defects. In this study, a new membrane is expected to improve the mechanical properties, as well as increase its antibacterial properties and reduce the cost while meeting the technical conditions of guided regeneration. Polylactic acid-glycolic acid/antibacterial decapeptide KSL-W (PLGA/KSL-W) shell-core structure antibacterial nanofiber membrane was prepared by coaxial electrospinning technology, and its microstructure was observed by SEM and TEM. Then, we tested the KSL-W drug sustained-release properties, mechanical properties, biocompatibility and effects on the proliferation and differentiation of osteoblasts MC3T3-E1 by in vitro experiments. In addition, we also tested its bone defect repair ability by constructing a rabbit model of apical periodontitis bone defect in vivo. The results show that the prepared nanofiber membrane has good microstructure and mechanical properties, and its sustained release solution has obvious antibacterial effect. Moreover, no matter one month or two months later, the in vivo bone defect repair effect of the nanofiber membrane group could reach a similar effect as that of the Bio-Gide group, but the number of inflammatory cells was significantly reduced. KSL-W (PLGA/KSL-W) shell-core structure antibacterial nanofiber membrane provides a new option for the repair and treatment of apical periodontitis bone defects.
