Construction Of Core-Crosslinked Star Polymer In Osteoporotic Mice Experiment
To investigate the role and underlying mechanism of LA-CCS/TEMPO in therapeutic application in osteoporotic mice with related bone diseases Ovariectomy model was induced in 8-week-old female WT mice via abdominal incision made to remove ovaries. After 9 weeks, alveolar bone resorption was quantified using micro-computed tomography and hematoxylin-eosin staining. Bbone marrow-derived macrophages were treated with macrophage colony-stimulating factor receptor activator of nuclear factor-κB ligand. The differentiation of osteoclasts was evaluated using tartrate-resistant acid phosphatase (TRAP) staining, which identifies TRAP-positive multinucleated cells indicative of mature osteoclasts. After 3 days castration surgery, each experimental group received intraperitoneal injections (0.5 ml/3 days) continuously for 9 weeks LA (dose: 50 μM), CCS/TEMPO (dose: 500 μM), and LA-CCS/TEMPO (dose: 50/500 μM). Micro-CT and HE analysis of trabecular bone volume percentage (percent bone volume, BV/TV) and bone mineral density (BMD) in the alveolar bone of ovariectomy mice. The OVX+Lig group exhibited significant alveolar bone resorption, with osteoclast counts significantly higher than those in the Sham+Lig group or OVX group alone. Notably, the LA-CCS/TEMPO group demonstrated comparable therapeutic efficacy to the Aln group in suppressing alveolar bone resorption compared to the OVX+Lig group. This study delineates the regulatory role of LA-CCS/TEMPO in regulating osteoclast differentiation and formation, which helps understanding the therapeutic efficacy in osteoporosis, periodontitis, and osteoporosis related periodontitis conditions.
