Domestic Material Vs Iroot Bp Plus On Hdpscs In Vitro
Pulp capping materials play an essential role in the treatment of living pulp preservation and are a subject of considerable interest in the study of endodontic diseases in young permanent teeth[1]. This study aims to compare the biological effects of iRoot BP Plus and novel domestic calcium silicate-based materials on human dental pulp stem cells (hDPSCs), providing theoretical basis for developing novel pulp capping agents. [1] da Rosa WLO, Piva E, da Silva AF. Disclosing the physiology of pulp tissue for vital pulp therapy. Int Endod J. 2018 Aug;51(8):829-846. With ethics committee approval, primary hDPSCs were isolated from third molars of 18-24-year-old volunteers (n=15) and authenticated through flow cytometry. Cells were exposed to material eluents for cytotoxicity (CCK-8 assay), migration (scratch assay), and osteogenic differentiation analyses. Molecular mechanisms were investigated using ALP staining, qRT-PCR, Western blot, and RNA-seq. Statistical significance was determined by ANOVA with p<0.05 threshold. Both materials demonstrated non-cytotoxicity and enhanced hDPSC migration. iRoot BP Plus exhibited superior upregulation of RUNX2 and ALP activity. RNA-seq revealed material-specific osteogenic regulation: iRoot BP Plus significantly increased POSTN and SOST expression compared to iMTA BP. While both bioceramics promote hDPSC osteogenesis through distinct pathways, iRoot BP Plus shows enhanced mineralization potential via POSTN/SOST modulation. These findings advance understanding of bioactive material-cell interactions, informing clinical material selection and next generation pulp capping agent development.
