Pharmacodynamic Substance Of Modified Zengshengping For Chemoprevention Of OSCC
To explore the pharmacodynamic substanc of Modified Zengshengping (ZSP-M) for chemoprevention of oral squamous cell carcinoma (OSCC). Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry (UPLC-MS/MS) was used to identify the components of ZSP-M entering blood and saliva. Molecular docking was performed between the possible bioactive ingredients and oral cancer-related targets in AutoDock Vina 1.1.2. In vitro experiments were conducted to investigate the proliferative effects of pharmacodynamic components on HOK, Cal27, SCC25 cells, mouse splenic lymphocytes, and their inhibitory effects on LPS induced NO release from RAW264.7. A total of 702 compounds were identified, including 20 blood primary prototype components and 31 saliva primary prototype components. 5 pharmacodynamic substance were selected: Scutellarin (SC), Chlorogenic Acid (CGA), Vitexin 2′'-O-rhamnoside (VOR), Maackiain (Ma), and Trifolirhizin (Tri). Low to medium concentrations of MA, SC, TRI, and high concentrations of VOR and CGA have non-toxic effects on HOK cells, but can inhibit the proliferation of Cal27 and SCC25 cells, suppress NO release, and promote the proliferation of mouse spleen lymphocytes. The experiment screened five pharmacodynamic substance from the Modified Zeng-sheng-ping, namely MA, SC, CGA, VOR, TRI. It is the key to the efficacy of drugs and has potential value for chemoprevention of oral cancer.
