Comparative Osteogenic Differentiation Of BMSC By Exosomes Of Different Origin
The difference of osteogenic differentiation ability of BMSCs induced by exosomes from different sources was compared in the inflammatory microenvironment. Mouse bone marrow mesenchymal stem cells were divided into 7 groups; The inflammation model was constructed with 1ug/ml LPS. Group A: adding osteogenic induction medium, group B: adding osteogenic induction medium after inflammation modeling. Group C: inflammatory model +BMSCs-EXO group; Group D: inflammatory model +BMSCs-EXO group after osteogenesis induction; group E: inflammatory model +M0-EXO group; Group F: inflammatory model +M2-EXO group. Group G: inflammatory model + BMSCs-EXO+M2-EXO group after osteogenesis induction. The osteogenic differentiation ability of bone marrow mesenchymal stem cells in each group was detected by alizarin red and alkaline phosphatase staining. The expressions of RUNt-related transcription factor 2, osteopontin, OSX and wnt pathway-related protein β-catenin in mouse bone marrow mesenchymal stem cells were detected by qRT-PCR and western blot. Compared with other groups, the expression levels of RUNt-related transcription factor 2, osteopontin and OSX in BMSCs-EXO+M2-EXO group after inflammation model + osteogenesis induction were significantly increased (P < 0.05). The expressions of wnt pathway-related proteins wnt1 and β-catenin were significantly increased (P < 0.05). The results show that in the inflammatory microenvironment, the bone marrow mesenchymal stem cell derived exosomes and M2-type macrophage derived exosomes after osteogenic induction of mouse bone marrow have better osteogenic differentiation ability.
