Expression And Mechanism Of Slc7a2 In Tongue Squamous Cell Carcinoma
To investigate how SLC7A2 affects cisplatin resistance in tongue squamous cell cells, and to reveal its specific role in the resistance mechanism. This will provide a new target for the treatment of tongue squamous cell carcinoma. We selected SLC7A2 from RNA-Seq of Cisplatin-resistant A549 cells, and then performed multi-omics combination analysis, cell line construction, RT-qPCR, and CCK8 to investigate the effects of SLC7A2. We analyzed RNA-seq in Cisplatin-resistant CAL27 cells and found that SLC7A2 expression in OSCC was down-regulated. Lower SLC7A2 expression is associated with poor prognosis in OSCC. SLC7A2 overexpression enhanced the repair ability of OSCC DNA and its insensitivity to cisplatin. Combined with multi-omics sequencing, activation of PRPP upregates the expression of SLC7A2 and enhances the sensitivity of OSCC cells to anti-tumor drugs. In addition, purine metabolic pathways suggest that SLC7A2 may be involved in the development of cisplatin resistance by regulating nucleotide supply, DNA repair capacity, and metabolic homeostasis. SLC7A2 may act as a tumor suppressor to regulate drug sensitivity, DNA repair ability, and cell proliferation in patients with OSCC.
