Fluorinated-Poss Resin: Microenvironment-Response And Anticaries Mechanism
This study addresses key challenges in caries prevention, including the insufficient synergy between antibacterial and remineralization effects and the limited mechanical durability of resin. Through molecular design, a fluorinated polyhedral oligomeric silsesquioxane (F-POSS) functionalized composite resin system was developed, systematically elucidating its microenvironment-responsive anti-caries mechanism. An innovative silica-based drug-loaded microsphere (ACP NPs@SiO₂) was designed to encapsulate pH- and light-responsive amorphous calcium phosphate nanoparticles (ACP NPs). These ACP NPs co-deliver metronidazole and fluoride ions, enabling precise drug release and sustained fluoride ion release in the acidic microenvironment of cariogenic biofilms. Surface silanization modification and graded particle filling techniques were employed to optimize the silica microspheres, thereby enhancing the mechanical properties and wear resistance of the resin. The incorporation of F-POSS facilitated the formation of a 3D crosslinked network, imparting hydrophobicity to the material, effectively reducing bacterial and protein adhesion. Its rigid cage-like structure covalently crosslinked with the resin matrix, thereby improving water resistance and stability. In vitro experiments confirmed that the material exhibits long-lasting antibacterial activity in a simulated oral environment, demonstrates good biocompatibility, and promotes the remineralization of demineralized dentin. This study provides new insights and strategies for the design of functionalized anti-caries resins and the prevention of dental caries.
