Minimalist Flexible Nanocapsules For Enhancement Of Dc-Mediated Tumor Immunotherapy

Traditional nanovaccines in dendritic cell-mediated immunotherapy are often limited by the risks associated with nanomaterials as carriers. Therefore, based on the "carrier free" strategy, this study simulated the extracellular vesicle structure to synthesize a novel biomimetic antigen nanocapsule (OVAnano) to enhance the efficacy of tumor immunotherapy. Preparation of biomimetic antigen nanocapsules simulating flexible extracellular vesicle structures using core assisted protein coating and selective etching methods. We have successfully prepared biomimetic antigen nanocapsules OVAnano that can simulate the structure of extracellular vesicles. OVAnano, with its flexible biomimetic structure, has good biocompatibility and efficiently promotes the maturation of DCs by activating the NF - κ B signaling pathway. OVAnano has highly purified antigens and exhibits significant antigen specificity, inducing CD8+and CD4+T cells to target specific antigens through DCs, thereby triggering strong anti-tumor immune responses. These findings indicate that our use of biomimetic antigen nanocapsules as a simple and effective strategy can effectively trigger strong immune responses against tumors, demonstrating its enormous potential in DCs mediated tumor immunotherapy and providing a new direction and strategy for future clinical applications.
