Multifunctional Micelles Based On Nanoenzymes For Periodontitis Therapy

The aim of this study was to construct a ROS-responsive micellar system loaded with cerium oxide nanozyme, which could be used as a multi-functional nanomaterial for bone regeneration, oxidative stress relief and antibacterial in the treatment of periodontitis. HA-PMs@Ce was synthesized via the self-assembly of PLLA-b-PLys/PBA and TA in deionized water containing CeO2, followed by coated with HA. Biocompatibility, ROS clearance, osteogenic efficacy, antibacterial activity and immune regulation effect of HA-PMs@Ce were tested. Bone formation and ROS clearance of HA-PMs@Ce were evaluated in SD rats with periodontitis bone defects induced by silk ligation. HA-PMs@Ce could quickly release TA and CeOx under high ROS microenvironment, showing stronger bactericidal activity. Meanwhile, HA-PMs@Ce exhibited excellent biocompatibility, promoted stem cell differentiation and reduced oxidative damage. In addition, HA-PMs@Ce improved oxidative stress by intervening the function of damaged mitochondrial organelles, reducing oxidative damage of mitochondria and treating ROS-related inflammatiom. HA-PMs@Ce showed specific recognition of inflammatory microenvironment, alleviating oxidative stress, promoting bone regeneration, and immune regulation of macrophage polarization, providing a new strategy for treating periodontitis.
