Multimodal Zif-8 Composite Microneedle Patch For Treatment Of Periodontitis

Periodontal disease is a chronic inflammatory and destructive disease. A topical drug delivery system is one of the non-surgical treatment strategies for periodontitis, but faces the dilemma of drug loss from periodontal pockets due to salivary flushing and muscle movement. Soluble microneedles (MN) can serve as a painless and efficient drug delivery system to address this issue. Therefore, In this study, different metal ions (Cu2+, Ag+, Mn2+) were doped into ZIF-8 nanoparticles with Gelatin Methacryloyl(GelMA)/Polyvinyl Alcohol(PVA)to construct a biodegradable microneedle patch for integrated antimicrobial and anti-inflammatory treatment of periodontal disease. A series of characterization tests (such as X-ray diffraction, scanning electron microscopy, and axial compression experiments) will be conducted to analyze the ZIF-8 composite microneedle patches, and its slow-release performance was tested by in vitro degradation experiments. The biocompatibility, anti-inflammatory and antibacterial properties of the composite microneedle patch were examined by the hemolysis rate, inflammatory cell counts and colony counts in SD rats. The results showed that the ZIF-8 nanoparticle variant was successfully synthesized and the composite microneedle patch had good morphology and mechanical penetration when the concentration of GelMA was 20%. 50 μg/ml of Cu-ZIF-8 reduced inflammatory cell counts in SD rats, and the hemolysis rate was below 5%, indicating good biocompatibility. In conclusion, the ZIF-8 composite microneedle patch has good antibacterial and anti-inflammatory activity and has potential in the treatment of periodontitis.
