Temporal And Spatial Changes Of GDNF

To investigate the temporal and spatial dynamics of various neurotrophic factors, with a focus on the role of glial cell line-derived neurotrophic factor (GDNF), in the repair process following mandibular injury in mice. A mouse mandibular injury model was established to investigate the temporal expression patterns of peripheral neurotrophic factors GDNF, NGF, and BDNF using qRT-PCR and immunofluorescence (IF) at 0 h, 12 h, 48 h, and 7 d post-injury. The results of quantitative real-time polymerase chain reaction (qRT-PCR) demonstrated that the expression levels of GDNF, NGF, and BDNF in the caudate putamen (CPU), prefrontal cortex (PFC), hippocampus (HIP), and cerebral cortex (CTX) were all elevated 12 hours post-mandibular injury, with GDNF showing the most significant increase in the CTX. At 12 h and 48 h after mandibular injury, the expressions of GDNF, BDNF, NGF, H2-D1 and H2-T23 in cpu, pfc, hip, ctx and the bone tissue around the injured side were increased to varying degrees, and the astrocytes were significantly activated and secreted GDNF. However, they all fell back to the initial level 7 days after injury.
