Ultrafine Platinum-Iridium Distorted Nanowires As Antimicrobial And Antioxidant In
In this work, ultrafine platinum-iridium distorted nanowires (PtxIr DNWs) were designed and synthesized to examine bactericidal and ROS-scavenging properties in the periodontal microenvironment. Additionally, the mechanisms of therapeutic effects on periodontitis were investigated. PtxIr DNWs were synthesized via a wet-chemical method, whose morphology and structure were characterized by TEM, XRD, and XPS. Enzyme catalytic activities were evaluated using UV-Vis spectrophotometry and oximetry. Flow cytometry and confocal microscopy were used to study cellular uptake and transport of the material. Cellular immunofluorescence detected inflammatory factor expression levels. Antibacterial activity was evaluated using MIC, live/dead staining, and SEM. The inhibitory effect was evaluated by imaging and histological staining. Microbial communities were analyzed by 16S rDNA sequencing. 1.The nanowires are 30±2.5 nm long and 1.7±0.5 nm in diameter, which comprises an alloy phase with irregular arranged atoms. UV-Vis spectrophotometry indicates that Pt2Ir demonstrated superior CAT and SOD enzyme activities compared to other tested materials. 2.Pt2Ir DNWs suppress LPS-induced ROS and iNOS overproduction. Antibacterial assays, biofilm staining, and SEM show they exhibit strong antibacterial and anti-biofilm activity. 3.Imaging and histological staining show Pt2Ir inhibit alveolar bone resorption and reduced inflammation.Harmful bacteria decreased significantly after treatment. This work developed ultrafine PtxIr DNWs to address bacterial infections and high ROS levels in periodontitis. The alloying and strain effects-induced distorted structure features surface defects and active atoms, providing CAT- and SOD-like activities. These show potential for clinical periodontitis management.
