Lycium Barbarum Polysaccharide Loaded Ros-Responsive Hydrogel For Ornj Treatment
osteoradionecrosis of the jaw (ORNJ) is associated with the overproduction of reactive oxygen species, and the current treatment for this disease is not satisfactory. Hydrogels were prepared by emulsion method and Thioketal bond. The preparation of hydrogel was verified by nuclear magnetic H spectrum, FTIR and SEM. The radiation-damaged cell model was established. To investigate the effects of hydrogel on cell proliferation and differentiation, angiogenesis, osteogenesis and metabolism. Crosstalk between HUVECs and BMSCs was verified using conditioned media. Transcriptome sequencing of HUVECs was performed to explore the mechanism of hydrogel action. The ORNJ rat model was established to verify the effects of hydrogel on antioxidation, promoting angiogenesis and promoting bone function. ROS responsive hydrogels were prepared successfully, in which Thioketal bonds were broken and LBPs were released in response to high ROS environment. Cell experiments showed that hydrogel had good biocompatibility and can enhance the proliferation and angiogenesis capacity of HUVECs, enhance cell metabolism, and promote glycolysis, after radiation damage. The metabolites released by HUVECs can induce histone lactation of BMSCs, activate osteogenic genes, and promote osteogenic differentiation, which has been verified in later animal experiments. We successfully prepared ROS responsive hydrogels loaded with LBPs, and verified that they have good mechanical property, can release LBP in response to high ROS environment, promote HUVECs proliferation and angiogenesis, enhance cell energy metabolism, induce histone lactation of BMSCs, activate osteogenic genes, and promote osteogenic differentiation. This has potential implications for the treatment of ORNJ.
