Nanosilicates Enhanced Pdlscs Osteogenesis Via Activation Of Wnt/Β-Catenin Signaling
To explore the effects of nSi on PDLSCs-mediated periodontal tissue regeneration and investigate the potential involvement of Wnt/β-catenin signaling pathway. The osteogenic differentiation of PDLSCs induced by nSi was evaluated through alkaline phosphatase (ALP) activity, alizarin red staining (ARS), and real-time quantitative PCR (qPCR) analysis. Additionally, the impact of Wnt/β-catenin signaling pathway in PDLSCs was investigated using Western Blot (WB), Immunofluorescent staining(IF). Moreover, IWR-1 was used to inhibit Wnt/β-catenin signaling pathway. 1. nSi enhanced osteogenic differentiation of PDLSCs. 2.Ingested nSi promoted the Wnt/β-catenin signaling pathway in PDLSCs. 3.IWR-1 treatment significantly downregulated key Wnt/β-catenin markers, reduced mineralized nodule formation, suppressed ALPL expression, and inhibited osteogenic differentiation genes and proteins. This study demonstrated that nSi promoted the osteogenic differentiation of PDLSCs, and nSi promoted this differentiation may through activation of the Wnt/β-catenin signaling pathway.
