Antibacterial Activity Of Cyclocarya Paliurus Polysaccharide On Oral Pathogens

With the increasing severity of antibiotic resistance, the antimicrobial properties of natural plant extracts have attracted widespread attention. This study investigates the inhibitory effects of Cyclocarya paliurus polysaccharide (CP) on Streptococcus mutans (S. mutans) and Porphyromonas gingivalis (P. gingivalis),aiming to explore its potential as a novel preventive agent for dental caries and periodontitis. The minimum inhibitory concentration (MIC) of CP against S. mutans and P. gingivalis was determined using the broth microdilution method. The biofilm formation and metabolic activity were assessed via crystal violet staining and MTT assays, respectively, at concentrations of 1/2×MIC, MIC, and 2×MIC. The effects on bacterial cell morphology were observed using scanning electron microscopy (SEM). CP exhibited MIC values of 0.25 mg/mL for S. mutans and 1 mg/mL for P. gingivalis. Biofilm formation and metabolic activity of both bacteria were significantly inhibited in a dose-dependent manner (P < 0.05). SEM revealed structural damage, including surface roughness, shrinkage, and pore formation in bacterial cells treated with CP. CP effectively inhibits the growth, biofilm formation, and metabolic activity of S. mutans and P. gingivalis, causing significant structural damage to bacterial cells. These findings suggest that CP has potential as a novel natural antimicrobial agent for preventing oral diseases.
