Application Of Adm Composite Piezoelectric Membrane In GBR
To design a novel bilayer guided bone regeneration (GBR) membrane composed of acellular dermal matrix (ADM) and piezoelectric layer poly L-lactic acid-chitosan-polydopamine (PLLA-CS-PDA), investigate its effects on the proliferation and differentiation of bone marrow mesenchymal stem cells (BMSCs) and vascular endothelial cells (VECs), and to provide experimental basis for the clinical application of bilayer composite membrane in repairing bone tissue defects. Fetal bovine skin was decellularized to get ADM, which was used as a soft tissue side barrier membrane. Poly L-lactic acid (PLLA) and chitosan (CS) were fabricated by electrospinning method as a guide membrane for hard tissue side. The spun membrane was modified with polydopamine (PDA) and then composite with ADM to prepare a bilayer piezoelectric composite membrane (PLLA-CS-PDA-ADM). The morphology, hydrophilicity and piezoelectric properties of PLLA-CS-PDA-ADM were observed. BMSCs and VECs were isolated and cultured, and the histocompatibility, osteogenic and angiogenic properties of the materials were detected. The two sides of the PLLA-CS-PDA-ADM bilayer composite membrane showed different surface characteristics under the SEM. The GBR membrane had excellent hydrophilicity, piezoelectric properties and biocompatibility. Under the piezoelectric drive provided by ultrasound, the GBR membrane effectively enhanced the adhesion, migration and osteogenic differentiation of BMSCs and the migration and tube formation of VECs. In this study, we successfully fabricated a novel bilayer piezoelectric composite membrane material, which shows enormous potential in osteogenesis and angiogenesis.
