Trxr-Mimetic Alendronate-Based Carbon Dot Nanozymes For Osteoporosis Treatment
To construct alendronate carbon dot nanozyme (Se-S/ALEN CDs) with the biomimetic activity of thioredoxin reductase, and to explore its therapeutic effect on osteoporosis. Se-S/ALEN CDs were synthesized by the hydrothermal method, and their physical and chemical properties were characterized. Methods such as CCK8 assay, RT-PCR, TRAP staining, and ALP activity detection were used to detect the effects of Se-S/ALEN CDs in anti-inflammation, promotion of osteogenic differentiation, promotion of angiogenesis, and inhibition of osteoclast differentiation in vitro. An ovariectomized mouse model of osteoporosis was adopted, and techniques such as Micro-CT and histological staining were used to evaluate the therapeutic effect on osteoporosis. Alendronate carbon dot nanozyme with the biomimetic activity of thioredoxin reductase was synthesized. In vitro studies showed that Se-S/ALEN CDs could significantly promote the proliferation and differentiation of osteoblasts, promote angiogenesis, and inhibit the differentiation of osteoclasts. In vivo results indicated that Se-S/ALEN CDs could effectively treat osteoporosis. This study demonstrates that alendronate carbon dot nanozyme with the biomimetic activity of thioredoxin reductase has a remarkable therapeutic effect on osteoporosis and is expected to serve as a nanozyme drug for the clinical treatment of osteoporosis.
