Manganese-Containing Bioglass Microneedles For Prevention The Oral Leukoplakia Carcinogenesis
Oral leukoplakia(OLK) is the most common oral potentially malignant lesion. In the present study, we developed a microneedle patches system(MN) containing manganese bioactive glass (MnBG-MN), which is aimed to provide an effective method for the treatment and carcinogenesis prevention of OLK. Manganese-containing bioactive glass (MnBG) were prepared by sol-gel method, and MN incorporating MnBG was designed.First, the surface microstructure, chemical structure, and ion release assay of MnBG were comprehensively characterized through Scanning Electron Microscopy (SEM),Fourier Transform Infrared Spectroscopy (FTIR), and Inductively Coupled Plasma Optical Emission Spectrometry (ICP-OES).Subsequently, the regulatory effects of MnBG-MN on cellular behaviors were systematically investigated in vitro. An animal model was established to evaluate the therapeutic and inhibitory effects of MnBG-MN on OLK. These effects were validated through comprehensive analyses of changes in animal body weight, Hematoxylin and Eosin (H&E) staining, immunohistochemical staining, and in vivo toxicity experiments. The MnBG-MN was successfully constructed. Preliminary findings indicated that Mn-BG could inhibit the growth of tumor cells but showed no significant inhibitory effect on normal cells. In vivo experiments disclosed that MnBG-MN exerted therapeutic impacts on OLK, with no obvious toxicity observed. Mn-BG effectively suppresses the proliferation of cancer cells. When combined with the MN, it enables precise targeting of OLK lesions. The MnBG-MN demonstrates excellent therapeutic and inhibitory efficacy against OLK. The integration of MnBG with MN technology offers a novel strategy for localized drug therapy in the treatment of OLK.
