Application Of Prp In Quiescence-Activation Dpscs
This study aims to analyze the key active components in PRP that regulate rest-activation. By detecting its impact on osteogenic differentiation of cells, this study provides new ideas for the in vitro culture of dental pulp stem cells and their application in bone tissue regeneration. RT-qPCR,CCK-8,EdU, ALP and ARS stainingstaining,Flow cytometry After 48-hour quiescence pretreatment of dental pulp stem cells, quiescent dental pulp stem cells were successfully obtained. Four different additives were used to activate the quiescent dental pulp stem cells. After 48 hours of activation, except for the Re + HPL group, the other three activation groups completely exited the quiescence period, and the proliferation ability of these three groups of cells was significantly increased compared with the blank control group.Transcriptomic analysis showed that compared with the Re + FBS group, the differentially expressed genes in the other activation groups were significantly enriched in pathways such as DNA replication, cell cycle, osteogenic differentiation, bone mineralization, and cellular senescence. Osteogenic induction results showed that the Re + PRP group exhibited the strongest osteogenic ability, followed by the Re + S-DPSC group and the Re + FBS group, while the Re + HPL group showed a relatively weak osteogenic ability. Platelet-rich plasma can successfully activate resting dental pulp stem cells, while human platelet lysate cannot. After being reactivated by platelet-rich plasma, resting dental pulp stem cells exhibit stronger proliferation and osteogenic differentiation capabilities.
