1D Cof/Naringin Ointment Periodontitis Photodynamic Therapy
Currently, traditional mechanical debridement techniques and antibiotic treatments for periodontitis are less effective, necessitating an efficient treatment strategy . This study designed a double-layer drug delivery system (COF-Por-DPP@Naringin) by loading one-dimensional covalent organic framework (COF-Por-DPP) and naringin into polyethylene glycol ointment. In the periodontal microenvironment, the system releases COF-Por-DPP and naringin. COF-Por-DPP generates abundant reactive oxygen species (ROS) under 808 nm laser irradiation, killing bacteria via antimicrobial photodynamic therapy (aPDT), while naringin exerts antioxidant effects for inflammation repair. CCK-8 assay evaluated biocompatibility and safety. In vitro, antibacterial effects were assessed by bacterial live/dead staining, CFU counting, and SEM analysis. In vivo, the male BALB/c mouse periodontitis model (6-8 weeks old, animal ethics was approved) was established. Micro CT, H&E, Masson, and IHC staining verified the in vivo anti-inflammatory and immunomodulatory effects. In vitro, COF-Por-DPP @Naringin showed >99% antibacterial efficacy (P < 0.05) against Porphyromonas gingivalis and Fusobacterium nucleatum. In vivo, COF-Por-DPP @Naringin significantly reduced periodontal pocket depth and alveolar bone resorption, with decreased IL-6, IL-1β and TNF-α levels. COF - Por - DPP@Naringin can generate ROS under light for killing periodontal bacteria and enhance periodontitis treatment. Naringin, with strong antioxidant properties, can effectively treat periodontal inflammation. In - vivo periodontal inflammation treatment verified its aPDT effect and antioxidant capacity. An effective periodontitis treatment strategy was provided.
