Paeonia Veitchii Components As Antibacterial And Anti-Inflammatory Itracananl Medication

The research aim is to explore the antibacterial and anti-inflammatory effects of Paeonia Veitchii active components to evaluate their potential as bifunctional itracananl medication. Paeoniflorin, chlorogenic acid and proanthocyanidin were selected and their individual and combined antibacterial and antibiofilm effects on Enterococcus faecalis were determined by bacterial susceptibility assay, crystal violet staining and scanning electron microscope. The mechanisms of membrane disruption, metabolic inhibition and virulence inhibition were preliminarily explored by scanning electron microscope, BacTiter-Lumi kit and qRT-PCR. The anti-inflammatory effects on cytokines of macrophages were determined with ELISA and the mechanisms were preliminarily predicted by network pharmacology. Individual use of paeoniflorin, chlorogenic acid and proanthocyanidin could inhibit both planktonic E. faecalis growth and prevent biofilm formation at a little higher concentration, and pairwise combination showed more effective inhibition with about two-fold lower active concentrations. These active components impacted E. faecalis by disrupting bacterial membranes, inhibiting energy metabolism and downregulating the genes expression associated with bacterial adhesion, biofilm formation, dictate survival, tissue injuries and stress response. The Paeonia Veitchii components reduced the secretion of IL-6, IL-1β and TNF-α and network pharmacology suggested they could influence stimulus response and apoptotic process through PI3K-Akt signaling pathway and MAPK signaling pathway. The active components of Paeonia Veitchii, paeoniflorin, chlorogenic acid and proanthocyanidin are potential bifunctional antibacteial and anti-inflammatory itracananl medication. The enhanced efficacy of combined usage suggested a potential strategy to promote the bio-activity of herbal components for clinical use.
