Chronic Stress Promotes Emt In Oscc Via Tgf-Β1 Induction.
Exploring the Impact of Chronic Stress on Epithelial-Mesenchymal Transition (EMT) in Oral Squamous Cell Carcinoma (OSCC) and Its Underlying Mechanisms, with a Focus on the Role of Transforming Growth Factor Beta 1 (TGF-β1). Through in vivo animal models, a chronic restraint stress model and an OSCC tumor-bearing model were established. Techniques such as Western blot, real-time quantitative polymerase chain reaction (qPCR), and enzyme-linked immunosorbent assay (ELISA) were employed to measure the expression levels of TGF-β1 and its effects on EMT-related markers in OSCC cells. Chronic stress was found to significantly upregulate the expression of TGF-β1 in OSCC cells (P<0.05), and induce the EMT process, as evidenced by the downregulation of the epithelial marker E-cadherin and the upregulation of the mesenchymal markers N-cadherin and Vimentin (P<0.05). Furthermore, blocking the TGF-β1 signaling pathway significantly inhibited the chronic stress-induced EMT process (P<0.05). Chronic stress promotes the epithelial-mesenchymal transition in OSCC by inducing the expression of TGF-β1, a process that may provide a biological basis for the invasion and metastasis of oral squamous cell carcinoma.
