In Vitro Study Of Emdogain Combined With Gelma For Vital Pulp Therapy.
This study evaluated Emdogain-gelatin methacryloyl (GelMA) hydrogel's dual functions in promoting odontogenic differentiation and suppressing inflammation in dental pulp stem cells (DPSCs) through in vitro experiments, exploring its potential for vital pulp therapy (VPT). Emdogain-GelMA composites were synthesized and characterized via scanning electron microscopy (SEM), degradation/swelling assays, and biocompatibility tests. DPSCs were divided into experimental (composite), Emdogain-alone, and control groups. CCK-8 assessed proliferation, while RT-PCR/Western blot quantified odontogenic markers (DSPP, ALP, RUNX2) and inflammatory factors (IL-6, TNF-α, TGF-β1, IL-10). ELISA monitored amelogenin release kinetics. Experimental results demonstrated that the Emdogain-Gelma composite exhibits a uniform porous structure and controlled sustained-release capability of Emdogain, with excellent biocompatibility (cell viability >90%). The material’s degradation rate matched the tissue regeneration process, while significantly upregulating odontogenic-osteogenic markers (DSPP, ALP, RUNX2) and bidirectionally modulating inflammatory factors (suppressing pro-inflammatory factors such as IL-6 and TNF-α, while activating anti-inflammatory factors like TGF-β1 and IL-10). The Emdogain-GelMA scaffold synergistically enhances VPT efficacy by sustaining bioactive component release and optimizing regenerative microenvironments. It overcomes traditional VPT limitations through dual-action osteogenic/anti-inflammatory modulation, reducing infection risks and extending tooth longevity, thereby advancing pulp preservation strategies.
