Versican V0/V1 Is Essential For The Proliferation Of Mdpcs

To evaluate the effect of Versican V0/V1 isoforms on the proliferation of mouse dental papilla cells (mDPCs). Immunofluorescence staining was performed to observe the expression of GAG-β, GAG-α and EdU. mDPCs were isolated, identified and transfected with siRNA. The proliferation ability of mDPCs was investigated using the Cell Counting Kit-8 and EdU assay. Flow cytometry was used to detect the cell cycle and apoptosis of mDPCs. Mouse tooth germs from mandibular first molars were isolated from mice at embryonic day 16.5 and transfected with siRNA. The morphological changes of tooth germs were observed under the microscope, and the width, height and cusp height of tooth germs were measured. The proliferation of cultured tooth germs were detected by EdU assays. The mDPCs of the si-Versican V0/V1 group showed weaker proliferation ability compared with the si-NC group (P<0.01) and the cell cycle progression of mDPCs in the si-Versican V0/V1 group was inhibited and the apoptosis was increased (P<0.0001). The si-Versican V0/V1 group showed a significant decrease in the width, height and cusp height of tooth germs. The proliferation of tooth germs in the si-Versican V0/V1 group was inhibited. Versican V0/V1 promotes the proliferation of mDPCs and cultured tooth germs, and affects the growth of cultured tooth germs and the development of tooth cusp.
