Effects Of Cse On Alveolar Bone Resorption In Occlusal Trauma
Cystathionine γ-lyase (CSE) has a significant role on alveolar bone remodeling. However, effects of CSE on alveolar crest resorption in occlusal trauma has not been studied. The aim of this study is to investigate impacts of traumatic occlusion on alveolar bone resorption in mouse models as well as the engagement of CSE in this process. Eight-week-old C57 WT mice and CSE-/- mice, with 20 for each type, were used to establish occlusal trauma models for 0, 1, 2, and 3 weeks. The degree of the reduction in the alveolar-crest height between the first and second molars was observed using HE staining, and the distance between the cement-enamel junction and the alveolar crest was calculated. The number of osteoclasts in the alveolar crest was calculated after TRAP staining. In both WT and CSE-/- mice, the alveolar crest height decreased after occlusion. Reduction in the alveolar crest height was mild in WT mice at 1 week and was significant at 3 weeks. Accordingly, osteoclasts in the alveolar crest were observed in WT mice at 1 week and actively proliferative at 3 weeks. Interestingly, the reduction in the alveolar crest height was smaller alongside lower osteoclast count in CSE-/- mice than that in WT mice at the same time points. As the modeling time of occlusal trauma prolongs, the alveolar crest height decrease and number of osteoclasts increase. CSE knockdown can curb such impacts induced by occlusal trauma with delayed abnormalities in the alveolar crest height and suppressed osteoclastgenesis.
